Ipamorelin and CJC-1295 are both classified as growth hormone secretagogues (GHSs), compounds that stimulate the release of growth hormone (GH), though they differ in structure and pharmacokinetics. Ipamorelin is a synthetic pentapeptide that primarily acts on the growth hormone secretagogue receptor (GHS-R1a) in the hypothalamus and pituitary, selectively promoting GH release without significantly affecting other pituitary hormones like prolactin. CJC-1295, a 29-amino-acid peptide, is a modified analog of GHRH (1–29) designed to mimic natural GH-releasing hormone and enhance pituitary GH secretion.
Overview
Both peptides stimulate the anterior pituitary, promoting natural, pulsatile GH release and subsequent increases in IGF-1, which mediates many of GH’s anabolic and metabolic effects.
CJC-1295 is a tetrasubstituted GHRH 1–29 peptide with four amino acid modifications at positions 2, 8, 15, and 27. These substitutions improve resistance to enzymatic degradation and allow partial binding to plasma proteins such as albumin, extending its half-life from approximately 10 minutes to 30 minutes. When equipped with a Drug Affinity Complex (DAC), CJC-1295 can remain in plasma for over 72 hours, while retaining strong affinity for GHRH receptors.
Ipamorelin is highly selective for GHS-R1a receptors and can trigger GH release rapidly, typically within the first two hours of administration. Unlike other GHSs, it has minimal impact on other pituitary hormones, making it a targeted GH stimulator.
When used in combination—often called a peptide stack—Ipamorelin provides the initial GH stimulation, while CJC-1295 maintains sustained GH release, producing complementary and prolonged effects on the GH–IGF-1 axis.
