Sermorelin is a 29-amino-acid synthetic peptide and the shortest fragment of growth hormone-releasing hormone (GHRH) capable of activating GHRH receptors. Structurally, it corresponds to GHRH (1–29) amide, retaining the functional region responsible for stimulating the pituitary gland. Sermorelin has been extensively studied in growth hormone deficiency models due to its ability to mimic the biological activity of full-length GHRH.

Overview

Sermorelin binds to GHRH receptors on the anterior pituitary, promoting the release of growth hormone (hGH). This stimulation may subsequently increase insulin-like growth factor-1 (IGF-1) levels, which mediate the anabolic effects of GH, supporting tissue growth, repair, and metabolism. Despite its shortened sequence, Sermorelin preserves the essential function of GHRH, enabling pulsatile GH release.

With an estimated half-life of 11–12 minutes, Sermorelin offers targeted receptor activity without significantly affecting other endocrine markers such as prolactin, insulin, cortisol, glucose, or thyroid hormones, highlighting its specificity and safety profile in growth hormone regulation.
